If gnometoaster lets you to edit an ISO, you can add files to the root of the CD, such as mydsl packages, backup file, and miscellaneous data files, but they will not be a part of the base system...at best they will be added after the system boots. This will allow you to add or replace files, but if you want to remove anything, or change the init process, you will need to remaster the KNOPPIX file, which i doubt can be done with gnometoaster.
