If your bios supports booting from usb then it is possible to boot directly from the usb-drive. No you don't have to make any partitions. Yes you can access the usb-drive (probably as sda1 or so). Don't remove the option "nousb" until you know what you are doing. This has to do with booting embedded in another os and depends on the hardware of the host computer. Your settings are saved using system-> backup/restore. Try it out and if something is unclear get back to the forum.
