If you are trying to boot from a USB flash drive another alternative is to try a different USB port.  I switched my flash drive from the "auxillary" USB ports on the front of my computer to an unpowered USB hub (keyboard in my case) and the "Can't find KNOPPIX" error went away and my embedded system booted.

I have yet to try it with a powered hub or using the USB ports directly on the motherboard (too lazy to crawl under my desk at the moment), but it may work from those as well.
